目的:研究前列腺素E2受体亚型EP1在不明原因复发性自然流产(URSA)中表达水平变化,分离鉴定子宫平滑肌细胞,并用特异性siRNA抑制EP1的表达,观察其对子宫平滑肌收缩率的影响,为抗RSA药物研究提供新的理论依据。方法:用荧光定量PCR检测两组子宫平滑肌细胞膜上EP1的表达分布情况;应用图像分析系统测量正常组和URSA组病人子宫平滑肌细胞收缩率;用特异性siRNA抑制URSA组子宫平滑肌细胞EP1的表达,分析其对在子宫平滑肌细胞收缩率的影响。结果:URSA组子宫平滑肌细胞中EP1的表达水平显著高于正常组(P<0.01),同时URSA组中子宫平滑肌细胞的收缩率显著高于正常组(P<0.01);EP1特异性siRNA和阴性对照siRNA处理URSA组子宫平滑肌细胞后,EP1特异性siRNA处理子宫平滑肌细胞收缩率较阴性对照siRNA处理组显著下降(P<0.05)。结论:在RSA病人中,外在刺激或内在因素导致EP1表达上调可能是促使子宫的异常收缩而发生流产的一个重要原因。因而PGE2/EP1系统可作为治疗RSA的一个潜在靶点。 |

Effect of EP1 on cell contraction in unexplained recurrent spontaneous abortion |

Abstract: |
Objective: The aim of the study was to characterize the role of EP1 in unexplained recurrent spontaneous abortion . Methods: A total of 48 induced aborters (Control Group ) and 37 recurrent spontaneous aborters(RSA Group) were enrolled. PGE2 receptor EP1 in MSMC at mRNA level was analysed using reverse transcription PCR and real-time PCR. Cell contraction was acquired with microscope analysis system in MSMC of URSA and Control group. EP1 siRNA or negative control siRNA treatment was operated in URSA MSMC. Results: In total,the level of EP1 was found increased in RSA group, consistently, cell contraction was found increased in RSA Group in comparison to controls. Knocdown of EP1 significantly decreased the level of cell contraction in URSA MSMC. Conclusions: Results suggest that upregulation of EP1 in unexplained recurrent spontaneous aborters, which probably mediates increased uterine contraction |
